Description:
The JES5-16E3 antibody reacts with mouse interleukin-10 (IL-10), an ~18 kDa cytokine also known as Cytokine Synthesis Inhibitory Factor (CSIF). IL-10 is primarily produced by Th2 cells, B1 cells, macrophages, and keratinocytes, and functions to inhibit Th1 cytokine synthesis and T cell proliferation while promoting Th2 responses.

Applications Reported:
Reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested by intracellular staining and flow cytometric analysis on stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
Recommended usage ≤ 0.25 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

Fluorochrome Information:
Alexa Fluor® 700 emits at 723 nm and is excited by the red laser (633 nm). Instruments typically require a 685 LP mirror and 710/20 filter.

Target Information

IL-10 encodes a cytokine produced mainly by monocytes and some lymphocytes. It regulates immune responses by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages, while enhancing B cell survival, proliferation, and antibody production. IL-10 also inhibits NF-kappa B activity and is involved in JAK-STAT signaling regulation. Mouse knockout studies indicate its essential role in intestinal immune regulation. Mutations in IL10 are linked to increased susceptibility to HIV-1 infection and rheumatoid arthritis.
Associated diseases include Graft-Versus-Host Disease and HIV Type 1 infection.
