Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells and binds glycosylated, fucosylated, sulfated sialylated glycoproteins such as CD34, GlyCAM-1, and MAdCAM-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes (≤ 0.2 µg/test).
A test is defined as the antibody amount staining cells in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test. Optimize titration for best performance.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01)

• Enhanced formulation for optimal staining with NovaFluor conjugates
• Add 5 µL per sample (regardless of NovaFluor count) before combining with cells
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells)

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor. It contains:

• C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Transmembrane and short cytoplasmic regions

Expressed on most leukocytes (except many memory T cells), monocytes, and polymorphonuclear cells.
