Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA).
CD45 is expressed on all hematopoietic cells except mature erythrocytes and platelets. It plays a key role in lymphocyte activation through tyrosine phosphatase activity.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse bone marrow cells at ≤0.4 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures with minimized spillover and spread.

Excitation: 552 nm; Emission: 700 nm; Laser: 561 nm (Yellow Laser)

Strain-specific Note:
NovaFluor Yellow 700 shows non-specific B cell staining in SJL mice, not observed in BALB/c or C57BL/6 strains.

Target Information

CD45 (Leukocyte Common Antigen, LCA)

• Receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins).
• Absent on non-hematopoietic tissues.
• Regulates Src-family kinases, T- and B-cell receptor signaling, cytokine receptor pathways, and integrin-mediated signal transduction.
• Exists as multiple isoforms due to alternative splicing; isoform expression varies by cell type and physiological state.
• Essential for immune cell signaling and survival regulation.
