Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sulfated sialylated glycoproteins such as CD34, glycam-1, and MAdCam-1, mediating lymphocyte rolling and homing to high endothelial venules (HEV).

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes at ≤ 0.2 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Careful titration recommended for optimal assay performance.

NovaFluor Compatibility:
Not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) required for optimal staining.
Add 5 µL per sample to antibody cocktails or single-color controls.

Technology:
NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor. It includes:

• C-type lectin domain
• EGF-like domain
• Two SCR sequences
• Spacer, transmembrane, and cytoplasmic regions

Expressed on all leukocyte classes except a portion of memory T cells, as well as on monocytes and polymorphonuclear cells.
