Target Information

Cas9 is an RNA-guided DNA endonuclease enzyme associated with the CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) adaptive immunity system in bacteria and genetic engineering.
The system is unique and flexible due to its dependence on RNA as the targeting element for nuclease to a desired DNA sequence.
Cas9 can be used for:

• Indel mutations, sequence replacements/insertions, and large deletions or genomic rearrangements
• Upregulation of endogenous genes
• Alteration of histone modifications or DNA methylation

Cas9 may be derived from Streptococcus pyogenes (SpCas9) or Staphylococcus aureus (SaCas9).
SaCas9 offers advantages such as smaller size, high efficiency, nickase activity, and specificity.
It recognizes an NNGRRT PAM and cleaves DNA at rates comparable to SpCas9 with optimal spacer lengths of 20–24 nucleotides.
SaCas9 (1053 AA) is smaller than SpCas9 (1368 AA) and suitable for all-in-one delivery of Cas9 and multiple gRNA cassettes using Adeno-associated vectors.
