전구체는 세포질에 존재하며 inflammasome 활성화 신호(ATP, bacterial flagellin 등)에 반응하여 절단되고 분비됩니다. 성숙형은 gasdermin-D(GSDMD) pore를 통해 세포 외로 방출됩니다. 분비는 단백질 변성에 의존하며 TMED10 cargo receptor에 의해 촉진됩니다.
Background
이 유전자가 암호화하는 단백질은 interleukin 1 cytokine family에 속합니다. 활성화된 대식세포에서 proprotein 형태로 생성되어 caspase 1(CASP1/ICE)에 의해 활성형으로 절단됩니다. 염증 반응의 주요 매개체로서 세포 증식, 분화, 세포사 등 다양한 세포 활동에 관여합니다. 중추신경계(CNS)에서 cyclooxygenase-2(PTGS2/COX2) 유도에 관여하여 염증성 통증 과민성을 유발합니다. 이 유전자와 다른 8개의 interleukin 1 family 유전자는 염색체 2에 위치한 cytokine gene cluster를 형성합니다.
