Target Information

COX2 converts arachidonate to prostaglandin H2 (PGH2), a key step in prostanoid synthesis including inflammatory prostaglandins. The conversion involves two reactions:

  1. Cyclooxygenase (COX) reaction — converts arachidonate to prostaglandin G2 (PGG2)
  2. Peroxidase reaction — reduces PGG2 to prostaglandin H2 (PGH2)

COX2 is constitutively expressed in some tissues such as endothelium, kidney, and brain, and is upregulated under pathological conditions like cancer and inflammation. Unlike PTGS1, which is ubiquitously expressed, COX2 upregulation is associated with increased cell adhesion, phenotypic changes, apoptosis resistance, and tumor angiogenesis.

In cancer cells, COX2 facilitates production of prostaglandin E2 (PGE2), influencing motility, proliferation, and apoptosis resistance. COX2 is naturally inhibited by calcitriol (active Vitamin D) and suppressed by glucocorticoids through transcriptional repression. It is a pharmacological target of NSAIDs such as aspirin, which reduce pain and inflammation mediated by COX2 activity.
