Target Information
COX2 converts arachidonate to prostaglandin H2 (PGH2), a key step in prostanoid synthesis including inflammatory prostaglandins. The conversion involves two reactions:
- Cyclooxygenase (COX) activity converting arachidonate to prostaglandin G2 (PGG2)
- Peroxidase activity reducing PGG2 to PGH2
COX2 is constitutively expressed in tissues such as endothelium, kidney, and brain, but is up-regulated under pathological conditions like cancer and inflammation. Up-regulation is linked to increased cell adhesion, phenotypic changes, resistance to apoptosis, and tumor angiogenesis. In cancer, COX2 promotes prostaglandin E2 (PGE2) production, influencing motility, proliferation, and apoptosis resistance. COX2 is inhibited by calcitriol (active Vitamin D) and targeted by NSAIDs such as aspirin.
