Product Specific Information

Description:
IM7 monoclonal antibody reacts with all isoforms of mouse CD44 (Pgp-1). CD44 is expressed by hematopoietic and non-hematopoietic cells. Bone marrow myeloid cells and memory T cells highly express this antigen, and peripheral B and T cells can upregulate CD44 expression. CD44 functions as an adhesion molecule through its binding to hyaluronate, an extracellular matrix component.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse bone marrow cells and splenocytes at ≤0.6 µg/test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Improved formulation over CellBlox
• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample before combining with cells
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells)

Target Information

CD44 is a 100 kDa type 1 transmembrane glycoprotein expressed on human leukocytes, brain white matter, and epithelial cells of the intestine and breast. It acts as a receptor for hyaluronic acid (HA), mediating cell-cell interactions, adhesion, and migration. CD44 is involved in lymphocyte activation, recirculation, and homing, and may be upregulated in certain carcinomas related to metastatic potential.

CD44 is expressed by hematopoietic and non-hematopoietic cells, epithelial tissues, and filopodia in cultured keratinocytes. Bone marrow myeloid cells and memory T cells show high expression. Alternative splicing generates multiple isoforms contributing to structural and functional diversity. CD44 dysfunction is associated with diseases such as superficial keratitis and lichen sclerosus, and may be linked to tumor metastasis formation.
