Product Specific Information

The IM7 monoclonal antibody reacts with all isoforms of mouse CD44 (Pgp-1). CD44 is expressed by hematopoietic and non-hematopoietic cells. Bone marrow myeloid cells and memory T cells highly express this antigen, and peripheral B and T cells can upregulate CD44 expression. CD44 functions as an adhesion molecule through binding to hyaluronate, an extracellular matrix component.

Each product contains:

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse bone marrow cells and splenocytes (≤0.6 µg/test).
A test is defined as staining a cell sample in 100 µL final volume (10⁵–10⁸ cells/test). Optimize antibody titration for best performance.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), featuring improved formulation over CellBlox. Required for optimal staining with NovaFluor conjugates.

• Add 5 µL per sample before combining with cells.
• For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD44 is a 100 kDa type I transmembrane glycoprotein expressed on human leukocytes, brain white matter, and various epithelial cells. It serves as a receptor for hyaluronic acid (HA) and mediates cell-cell interactions, adhesion, and migration. CD44 participates in lymphocyte activation, recirculation, and homing. Expression may increase in carcinomas, potentially linked to metastatic potential.
Alternative splicing of CD44 transcripts generates diverse isoforms. Diseases associated with CD44 dysfunction include superficial keratitis and lichen sclerosus. CD44 may also be implicated in tumor metastasis formation.
