Target Information

Proteolytic degradation is critical for maintaining proper levels of short-lived and regulatory proteins involved in metabolism, stress response, antigen presentation, receptor modulation, and cell cycle regulation.
The ubiquitin-proteasome pathway degrades most cytosolic and nuclear proteins, while others are processed via the vacuolar pathway (endosomes, lysosomes, ER).
The 26S proteasome (~2.5 MDa) comprises a 20S core and 19S regulatory subunits that recognize ubiquitinated proteins and facilitate their degradation.
Defects in this pathway are linked to diseases such as cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
