Target Information

This gene encodes a G protein-coupled receptor with selectivity for three chemokines: IP10 (interferon-γ-inducible 10 kDa protein), Mig (monokine induced by interferon-γ), and I-TAC (interferon-inducible T cell α-chemoattractant). These chemokines belong to the CXC subfamily, characterized by a single amino acid separating the first two conserved cysteines.

Binding of these chemokines to CXCR3 induces cellular responses involved in leukocyte trafficking, such as integrin activation, cytoskeletal rearrangement, and chemotactic migration. Inhibition by Bordetella pertussis toxin suggests coupling with Gi-type G proteins. Signal transduction may involve enzymes similar to those activated by other chemokine receptors.

Chemokine-induced cellular desensitization occurs via receptor phosphorylation and internalization, followed by recycling to the cell surface after dephosphorylation. CXCR3 is highly expressed in effector/memory T cells and in T cells within inflamed tissues. The chemokines IP10, Mig, and I-TAC are commonly produced at inflammation sites, implicating CXCR3 in inflammatory cell recruitment. Thus, CXCR3 is a potential target for small molecule antagonists aimed at treating inflammatory diseases. Multiple transcript variants encoding different isoforms have been identified.
