Target Information

The CXCR3 gene encodes a G protein-coupled receptor that binds chemokines IP10 (interferon-γ-inducible 10 kDa protein), Mig (monokine induced by interferon-γ), and I-TAC (interferon-inducible T-cell alpha chemoattractant). These chemokines belong to the CXC subfamily, characterized by a single amino acid separating the first two conserved cysteine residues.

Binding of chemokines to CXCR3 triggers cellular responses involved in leukocyte trafficking, including integrin activation, cytoskeletal rearrangement, and chemotactic migration. Pertussis toxin inhibition suggests coupling with Gi-type heterotrimeric G proteins. Signaling may involve cascades similar to other chemokine receptors.

Chemokine-induced desensitization leads to transient signaling due to receptor internalization, with responsiveness restored after receptor dephosphorylation and recycling. CXCR3 is highly expressed in effector/memory T cells and T cells within inflamed tissues. The receptor and its ligands play key roles in recruiting inflammatory cells, making CXCR3 a potential target for small-molecule antagonists in inflammatory disease treatment. Multiple transcript variants encoding different isoforms have been identified.
