Product Specific Information

The TMP4 monoclonal antibody reacts with human CD133 (Prominin-1), a 120 kDa member of the pentaspan family of proteins, including Prominin-2. These proteins are expressed in plasma membrane protrusions such as epithelial microvilli. CD133 exists in alternatively spliced isoforms and is glycosylated in a tissue-dependent manner. It was first identified on CD34+ hematopoietic progenitors and is widely used as a marker for stem cells and cancer stem cells, including those from colon, brain, and prostate cancers.

The TMP4 antibody recognizes a distinct epitope from the EMK08 clone, indicating non-overlapping binding sites.

Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended usage is 5 µL (0.25 µg) per test in 100 µL final volume.

Target Information

CD133 (AC133) is a 97 kDa glycoprotein with 5 transmembrane domains, appearing as a 120 kDa band due to glycosylation. It is expressed on immature hematopoietic stem cells but not mature blood cells. CD133 is also a marker for neural and embryonic stem cells and is expressed in certain cancers, including leukemias and brain tumors. Low-level expression is observed in kidney, pancreas, placenta, and fetal liver. Mutations in CD133 are linked to retinitis pigmentosa and Stargardt disease. Expression is regulated by multiple tissue-dependent promoters and alternative transcripts.
