Specificity Comments:
This antibody recognizes a protein of 65–76 kDa identified as antichymotrypsin (AACT). AACT is a plasma protease inhibitor synthesized in the liver as a single glycopeptide chain. In humans, normal serum levels are approximately one-tenth of plasma concentrations, which increase in response to trauma, surgery, or infection. Elevated AACT levels have been reported in cerebrospinal fluid and plasma of Alzheimer’s disease patients. PSA and its SDS-stable complex with AACT are used as markers for prostate cancer diagnosis. AACT deficiency may be associated with chronic liver disease. This antibody reacts with histiocytes and histiocytic neoplasms and is widely used to identify histiocytes and their tumor derivatives. Acinar tumors of the pancreas and salivary gland may also show AACT positivity.

Function: Alpha-1-ACT is an early-stage acute-phase plasma protein and a serine protease inhibitor that inactivates chymotrypsin, cathepsin G, and chymase. It is tightly associated with amyloid plaques in Alzheimer’s disease and aged brain tissue. Regulation of serine proteases and their inhibitors plays an important role in neuromuscular differentiation. PSA, a chymotrypsin-like serine protease, is predominantly complexed to Alpha-1-ACT.
