Target Information

Hyaluronic acid (HA) is a high molecular weight unbranched polysaccharide that forms part of the extracellular matrix in organisms ranging from bacteria to mammals. It consists of alternating glucuronic acid and N-acetylglucosamine residues linked by β-1,3 and β-1,4 glycosidic bonds. HA is synthesized at the plasma membrane by membrane-bound synthase and extruded into the extracellular space, serving roles in space filling, joint lubrication, and as a migration matrix for cells.

HA production increases during wound healing and tissue repair, supporting blood vessel and fibroblast ingrowth. Serum HA concentration changes are linked to inflammatory and degenerative conditions such as rheumatoid arthritis. The interaction between HA and the leukocyte receptor CD44 is critical for leukocyte homing, and overexpression of HA receptors correlates with tumor metastasis.

HAS1 is a member of the vertebrate gene family encoding hyaluronan synthases, showing significant homology to the hasA gene product of Streptococcus pyogenes.
