Product Specific Information
This STLALYV monoclonal antibody recognizes human survivin (Birc5), a member of the inhibitor of apoptosis (IAP) family. Survivin inhibits caspase activation, negatively regulating apoptosis, and plays a key role in mitosis and cytokinesis. It is highly expressed in tumors and fetal tissue but absent in terminally differentiated and non-proliferating cells.
Deletion studies in lymphocytes show survivin is essential for cell division but not for survival inhibition.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) according to BestProtocols®: Protocol B (nuclear intracellular proteins).
- Recommended use: 5 µL (0.5 µg) per test (100 µL final volume).
- Cell number: 10⁵–10⁸ cells/test (empirical).
- Excitation: 488–561 nm; Emission: 578 nm.
- Compatible lasers: Blue, Green, Yellow-Green.
- Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Survivin (IAP4) is the smallest member of the Inhibitors of Apoptosis Protein (IAP) family, involved in cell signaling, division, and metabolism. It exhibits high expression in cancers and fetal tissues but low in most adult tissues. Survivin contains a single baculovirus IAP repeat (BIR) domain and lacks a RING finger domain.
During mitosis, survivin associates with microtubules of the mitotic spindle; disruption of this interaction increases caspase-3 activity, promoting apoptosis. It is also expressed in brain tissues after injury, suggesting a role in neural cell proliferation. Multiple transcript variants exist for this gene.
