Target Information

Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of blood plasma proteins.
It inhibits amyloid fibril formation by APP, APOC2, B2M, CALCA, CSN3, SNCA, and aggregation-prone LYZ variants (in vitro) without requiring ATP.
Clusterin maintains partially unfolded proteins for subsequent refolding by other chaperones such as HSPA8/HSC70 but does not refold proteins itself.
Binding to cell surface receptors triggers internalization and degradation of chaperone-client complexes.
Secreted isoforms protect cells against apoptosis and complement-mediated cytolysis.
Intracellular isoforms interact with ubiquitin and E3 ligase complexes to promote proteasomal degradation of target proteins, modulating NF-kappa-B activity.
Nuclear isoforms promote apoptosis, while mitochondrial isoforms suppress BAX-dependent cytochrome c release and inhibit apoptosis.
Clusterin plays a key role in regulating cell proliferation.
