Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 upregulates stress response genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 contains an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is important for p53 activation and stabilization, while phosphorylation at Ser392 is associated with human tumor formation. p53 plays roles in blocking proliferation of damaged cells, acting as an anticancer agent, and is implicated in aging, oxidative stress, and learning and memory deficits. Normally expressed at low levels, p53 is upregulated in transformed cells and contributes to malignancy. Mutations in the TP53 gene are common in human cancers and can lead to loss of tumor suppressor activity, including in hereditary cancer syndromes such as Li-Fraumeni syndrome.
