Target Information
GRP78 is a 78 kDa glucose-regulated protein belonging to the HSP70 family. It resides in the endoplasmic reticulum (ER) and transiently associates with newly synthesized secretory and membrane proteins. GRP78 interacts with misfolded proteins, marking them for degradation. The conserved KDEL sequence at its C-terminus ensures ER retention via the KDEL receptor.
GRP78 plays a crucial role in protein folding, degradation of misfolded proteins, and maintenance of cell homeostasis. It prevents apoptosis and serves as a biomarker for hypoglycemia. GRP78 provides neuroprotection against oxidative stress and glutamate toxicity. Reduced GRP78 levels have been observed in Alzheimer’s disease and are associated with PS1 gene mutations. Induction of GRP78 is linked to drug resistance in cancer cells.
