Product Specific Information
The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2). Foxp3 is a 49–55 kDa transcriptional regulator involved in Treg cell function. The antibody epitope maps to amino acids 75–125 of mouse Foxp3. In humans, this region is alternatively spliced, whereas in mice it is not, indicating species-specific regulation.
FJK-16s is optimized for use with the Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523-00). Other fixation and staining buffers are not recommended.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Intracellular staining and flow cytometry of mouse splenocytes using the Foxp3/Transcription Factor Buffer Set.
Recommended Usage: ≤0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody titration recommended for optimal results.
Excitation: 488 nm

Target Information
FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is highly expressed in CD25⁺CD4⁺ regulatory T cells and absent in CD4⁻/CD8⁺ T cells. FOXP3 mutations cause the scurfy phenotype in mice and IPEX syndrome in humans. It is also regulated by p53 in response to DNA damage in human breast and colon cancer cells.
