Description:
The eBio4B10 monoclonal antibody reacts with mouse, rhesus monkey, and human T-bet. T-bet is a Th1-specific T-box transcription factor critical for Th1 CD4+ T cell lineage development. T-bet deficient mice show impaired Th1 cell development, while ectopic expression of T-bet skews differentiation toward Th1 lineage. T-bet expression is induced by IFN-γ and regulates IFN-γ expression through chromatin remodeling. It also regulates IL-12Rβ and IL-2 expression and plays a role in class-switch recombination in B cells.
Applications Reported:
This antibody is reported for use in intracellular staining followed by flow cytometric analysis.
Applications Tested:
Tested by intracellular staining and flow cytometric analysis of normal human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) and protocol.
Recommended usage ≤ 0.25 µg per test (100 µL final volume, 10⁵–10⁸ cells/test).
Antibody should be titrated for optimal performance.

Target Information
T-bet (TBX21) is a T-cell–specific transcription factor with a T-box DNA-binding domain. It controls expression of the Th1 cytokine IFN-γ and directs differentiation toward Th1 versus Th17 lineages. T-bet cooperates with Stat4 in programming chromatin architecture for Th1 gene expression. Genetic variations in T-bet are linked to aspirin-induced asthma and nasal polyps.
