Target Information
Atrophin-1 contains a polyglutamine repeat, expansion of which is responsible for dentatorubral and pallidoluysian atrophy. The protein encoded by this gene interacts with atrophin-1 and is a member of the NEDD4-like protein family. These proteins act as E3 ubiquitin-ligases, regulating trafficking decisions such as protein targeting to proteosomes or lysosomes. The encoded protein has four tandem WW domains and a HECT domain, functioning as a transcriptional corepressor of p45/NFE2 and possibly regulating immune responses via Notch-mediated signaling. It is highly similar to the mouse Itch protein, involved in erythroid and lymphoid cell differentiation.
