Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein belonging to the immunoglobulin family of receptors. It plays a crucial role in signal transduction, T cell differentiation, and activation. CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with T cell receptor (TCR) for MHC class I molecules.
CD8α is essential for MHC-I binding and is also found on subsets of T cells, NK cells, monocytes, and dendritic cells as disulfide-linked homodimers. Interaction with MHC-I/peptide complexes on antigen-presenting cells triggers recruitment of Lck kinase, leading to lymphokine production, motility, and activation of cytotoxic T lymphocytes (CTLs). Activated CTLs are critical for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
