• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

The protein encoded by the classic MBP gene is a major constituent of the myelin sheath of oligodendrocytes and Schwann cells in the nervous system. MBP-related transcripts are also present in bone marrow and the immune system. These mRNAs arise from the long MBP gene (also called Golli-MBP) that contains additional upstream exons.
Alternative splicing from the Golli and MBP transcription start sites produces two sets of MBP-related transcripts and gene products. Golli mRNAs encode hybrid proteins with N-terminal Golli sequence linked to MBP sequence, while the second family produces the classic myelin basic proteins. This complex gene structure is conserved among species, suggesting functional and regulatory importance.
