Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 forms a complex with CD132 (common gamma chain) to create the IL-7 receptor (IL-7R), a type I glycoprotein (~75–80 kDa). It is expressed by immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding triggers signal transduction through tyrosine kinase pathways including Jak/STAT. IL-7 is essential for lymphocyte development and T-cell homeostasis. CD127 down-regulation is characteristic of CD4+CD25+ regulatory T cells (Tregs), providing a more accurate marker for the Foxp3+ suppressive population.

Binding of eBioRDR5 to PBMCs can be blocked by pre-incubation with recombinant human IL-7.
Applications reported: Flow cytometric analysis.
Applications tested: Pre-diluted and validated on human peripheral blood cells at 5 µL (0.25 µg)/test.

Brilliant Violet™ 786 (BV786) emits at 786 nm and is suitable for cytometers with a 405 nm violet laser.

Usage Notes

• Use Super Bright Complete Staining Buffer or Brilliant Stain Buffer to minimize non-specific polymer interactions.
• Light-sensitive: protect from light.
• Fixation: compatible with IC Fixation Buffer or 1-step Fix/Lyse Solution; not compatible with methanol-based fixation.
• Excitation: 407 nm; Emission: 786 nm; Laser: Violet Laser.

Target Information

CD127 (Interleukin-7, IL-7) regulates lymphopoiesis. The IL-7 receptor is a heterodimer of alpha and gamma chains, mediating signal transduction and promoting proliferation of B cell precursors, thymocytes, and mature T cells. CD127 dysfunction is associated with severe combined immunodeficiency (SCID).
