Product Specific Information

The MIH18 monoclonal antibody reacts with human CD273 (PD-L2 or B7-DC), a member of the B7 family (~25 kDa) within the immunoglobulin superfamily. B7-DC is mainly expressed by dendritic cells and monocytes/macrophages and serves as a ligand for PD-1, not CD28/CTLA-4. The PD-1 and B7-DC/PD-L2 interaction can either stimulate or suppress T cell proliferation depending on activation state. MIH18 is a blocking antibody, useful for studying APC/T cell interactions and expression profiling of B7-DC/PD-L2 in humans.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry using human monocyte-derived dendritic cells. Use 5 µL (0.06 µg) per 100 µL test volume.
Excitation/Emission: 488 nm / 710 nm (Blue laser)
Recommended filter: 710/50 bandpass (695/40 acceptable alternative)
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (#00-8222-49) or 1-step Fix/Lyse Solution (#00-5333-54).
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Programmed death-ligand 2 (PD-L2, B7-DC) is a B7 family ligand that binds PD-1. It is predominantly expressed in dendritic cells but can be induced in various immune and non-immune cells. PD-L2 expression is upregulated by IL-4, TNF-α, and IFN-γ. PD-L2 has 2–6× higher affinity for PD-1 than PD-L1 and functions as an inhibitory checkpoint molecule regulating T cell activation and cytokine secretion. PD-L2 can also stimulate T cells independently of PD-1, suggesting additional receptors such as RGMb. Interaction between PD-L2 and RGMb contributes to respiratory tolerance regulation through BMP/neogenin pathways. The PD-L2 monomer consists of a 201-aa extracellular domain, 21-aa transmembrane domain, and 32-aa cytoplasmic domain.
