Target Information

In the intact cell, DNA closely associates with histones and other nuclear proteins to form chromatin. The remodeling of chromatin is a critical component of transcriptional regulation, and acetylation of nucleosomal histones plays a major role in this process. Acetylation of lysine residues in histone tails induces conformational changes, increasing DNA accessibility to transcription factors.

Several mammalian proteins act as histone acetylases, including GCN5, PCAF, p300/CBP, HAT1, and TAF II p250. Conversely, histone deacetylases (HDACs) are associated with transcriptional silencing. The HDAC family includes HDAC1–9. HDAC9 and HDAC9a are alternatively spliced isoforms; HDAC9a is 132 amino acids shorter but retains the catalytic domain and deacetylase activity. Both isoforms repress myocyte enhancer-binding factor 2-mediated transcription and are expressed in brain, skeletal muscle, kidney, placenta, and pancreas.
