Product Specific Information

  • Tested in direct ELISA (for coating use antibody at 1–5 µg/mL).
  • For Immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

ATG5 (Autophagy Related 5) is a key component of the autophagy pathway and may play an important role in apoptosis. It is involved in several cellular processes including:

  • Mitochondrial quality control after oxidative damage
  • Negative regulation of innate antiviral immune response
  • Lymphocyte development and proliferation
  • MHC II antigen presentation
  • Adipocyte differentiation

Following conjugation to ATG12, the ATG5–ATG12 complex participates in autophagosome formation and contributes to autophagic cell death by interacting with Fas-associated protein with death domain (FADD). The ATG5–ATG12 conjugate also associates with innate immune response proteins such as RIG-I and VISA (IPS-1), inhibiting type I interferon production and permitting viral replication.
Dysfunction of ATG5 is associated with diseases such as spinocerebellar ataxia.
