Product Specific Information
- Immunohistochemistry (PFA-fixed): Incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
ATG5 (Autophagy Related 5)는 자가포식(autophagy)에 필수적인 단백질로, 세포 사멸(apoptosis) 과정에서도 중요한 역할을 합니다.
또한 산화 손상 후 미토콘드리아 품질 조절, 선천적 항바이러스 면역 반응의 음성 조절, 림프구 발달 및 증식, MHC II 항원 제시, 그리고 지방세포 분화 등 다양한 세포 과정에 관여합니다.
ATG12와의 결합을 통해 오토파고좀 형성에 참여하며, FADD 단백질과의 상호작용을 통해 자가포식성 세포 사멸을 유도합니다.
ATG5-ATG12 복합체는 autophagosome 형성 직전 또는 직후 막으로부터 분리되는 컵 형태의 격리막(isolation membrane)을 형성합니다.
또한 RIG-I 및 VISA(IPS-1)와 같은 선천 면역 단백질과 결합하여 type I interferon 생성을 억제하고 바이러스 복제를 허용합니다.
ATG5 기능 이상은 소뇌실조증(spinocerebellar ataxia) 등과 관련이 있습니다.
