Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation.
p53 activation leads to transcription of specific genes such as p21waf1, regulating G1 or G2/M arrest. Structurally, p53 has an N-terminal transactivation domain, a central DNA-binding domain, an oligomerisation domain, and a C-terminal regulatory domain.

Phosphorylation at specific sites, including Ser15 and Ser37, is crucial for p53 activation and stabilization. p53 acts as a key anticancer factor by preventing proliferation of damaged cells. It is also associated with tumor formation (e.g., Ser392 phosphorylation), aging, oxidative stress, and cognitive functions such as long-term potentiation (LTP).

Mutations in TP53 are common in human cancers, leading to loss of DNA-binding and tumor suppressor activity. Germline mutations are also linked to cancer-prone syndromes such as Li-Fraumeni syndrome.
