Target Information

EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to various environmental stresses. PKR plays a key role in antiviral defense mediated through its activation by double-stranded RNA produced during viral infection. It is involved in IFN-induced innate antiviral responses, virus-induced apoptosis, and regulation of cell growth and differentiation.

PKR induces apoptosis by up-regulating Fas expression and mediates FADD/Caspase-8 death signaling. Upon viral infection, PKR acts as a dual regulator of both cell survival and death pathways through kinase-dependent and -independent mechanisms. It modulates multiple pathways, including NF-kappaB activation, p53, p38, and PDGF signaling. PKR has been implicated in tumor suppression and malignancy, as well as in neurodegenerative diseases such as Alzheimer’s, Huntington’s, and amyotrophic lateral sclerosis.
