Preparation:
Produced from sera of goats immunized with highly pure Recombinant Human TPO. Anti-Human TPO-specific antibody was purified by affinity chromatography employing an immobilized Human TPO matrix.

Sandwich ELISA:
To detect Human TPO by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity-purified antibody, in conjunction with PeproTech Biotinylated Anti-Human TPO (500-P49GBt) as a detection antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Human TPO.

Western Blot:
To detect Human TPO by Western Blot, this antibody can be used at a concentration of 0.1–0.2 µg/mL. Used with compatible secondary reagents, the detection limit for Recombinant Human TPO is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Thrombopoietin (TPO, THPO, MPL ligand, megakaryocyte colony-stimulating factor)
TPO is responsible for platelet generation through its action on hematopoietic cells committed to the megakaryocyte lineage. Binding of thrombopoietin to its receptor, MPL (CD110), initiates signaling via JAK/STAT, MAPK, and PI3K/AKT pathways, leading to megakaryocyte proliferation and differentiation and ultimately platelet production (thrombopoiesis).
Thrombopoietin is constitutively produced by the liver and has a predicted molecular weight of approximately 38 kDa.
