Product Specific Information

The eBio1D4B monoclonal antibody reacts with mouse CD107a, also known as lysosomal-associated membrane protein-1 (LAMP-1). CD107a is a type I lysosomal membrane protein that is extensively glycosylated and expressed in late endosomes-lysosomes of all cells. It is transiently expressed on the surface of degranulating cytolytic T cells and has been implicated in cancer metastasis and lysosomal storage disorders.

Applications Tested:

  • Validated for intracellular staining and flow cytometry using mouse thioglycolate-induced peritoneal exudate cells

Target Information

LAMP1 (CD107a), also known as lysosome-associated membrane protein-1, is a major component of the lysosomal membrane. Approximately 1–2% of total CD107a is found on the plasma membrane. It is a heavily glycosylated membrane protein with a signal peptide, 18 N-linked glycosylation sites, a single transmembrane domain, and a short cytosolic tail. LAMP1 and LAMP2 are involved in lysosome biogenesis and required for fusion of lysosomes with phagosomes.

Upon cell activation, LAMP1 translocates to the plasma membrane via a tyrosine-based motif (YXXI). Mutations in this motif disrupt lysosomal localization, resulting in cycling between plasma membrane and endosome. Surface LAMP1/2 promotes PBMC adhesion to vascular endothelium, suggesting a role in inflammation. Increased LAMP1 expression is observed in neurons and glial cells near senile plaques in Alzheimer’s disease and in macrophages during thymic involution. LAMP1 serves as a marker of mast cell activation.
