Product Specific Information
The monoclonal antibody 29A1.4 recognizes mouse NKp46 (CD335), a member of the natural cytotoxicity receptor (NCR) family. CD335 is a glycoprotein with two Ig-like domains and a short cytoplasmic tail, expressed specifically on NK cells, including immature NK cells (DX5⁻ CD3⁻), allowing discrimination between NKT and NK cells (NKp46⁺, CD3⁻).
Unlike many NK markers, staining with 29A1.4 is not strain-specific and has been shown in C57Bl/6, SJL, CBA/CA, and BALB/C strains. NKp46 plays a role in NK cell-mediated lysis of tumor and pathogen-infected cells.
The 29A1.4 antibody can activate NK cells in vitro but does not deplete them in vivo.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤1 µg/test. Optimal titration is recommended.

Target Information
NKp46 (CD335, NCR1) is a cytotoxicity-activating receptor contributing to the enhanced tumor cell lysis activity of activated NK cells. It consists of two Ig-like domains and is structurally related to leukocyte immunoglobulin-like (LIR) and killer inhibitory receptors (KIR).
