Product Specific Information

The monoclonal antibody 29A1.4 recognizes mouse NKp46 (CD335), a member of the natural cytotoxicity receptor (NCR) family. CD335 is a glycoprotein with two Ig-like domains and a short cytoplasmic tail, expressed uniquely on NK cells, including immature NK cells. It allows discrimination between NKT cells and NK cells (NKp46+, CD3-). Staining is not strain-specific and has been confirmed in C57Bl/6, SJL, CBA/CA, and BALB/C strains.

NKp46 plays a role in NK cell-mediated lysis of tumor and pathogen-infected cells. The 29A1.4 antibody activates NK cells in vitro but does not deplete them in vivo.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤1.0 µg/test (100 µL final volume). Cell count may range from 10^5–10^8 cells/test. Titrate for optimal performance.

Brilliant Violet™ 711 (BV711) emits at 713 nm and is suitable for cytometers with a 405 nm violet laser. Protect from light due to photo-induced oxidation sensitivity.

When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54). BV711 is not compatible with methanol-based fixation.
