Target Information

LATS2 is a serine/threonine protein kinase of the LATS tumor suppressor family. It interacts with p53 regulators and contributes to mitotic fidelity and genomic integrity. LATS2 is central to organ size control and tumor suppression by restricting proliferation and promoting apoptosis.

The LATS signaling pathway involves a kinase cascade where STK3/MST2 and STK4/MST1 activate LATS1/2, which phosphorylates and inactivates YAP1 and WWTR1/TAZ. Phosphorylation of YAP1 by LATS2 prevents nuclear translocation, regulating genes involved in cell proliferation, death, and migration. LATS2 also negatively regulates the G1/S transition by down-regulating cyclin E/CDK2 activity.
