Target Information
Calcineurin is a Ser/Thr, calcium and calmodulin-dependent protein phosphatase essential in T cell activation. It consists of two subunits:
- Calcineurin A (CnA): 60 kDa catalytic subunit
- Calcineurin B (CnB): 19 kDa regulatory subunit containing four EF-hand calcium binding sites
Calcineurin-mediated dephosphorylation of NF-AT is critical for its activation, nuclear translocation, and gene expression in T-cells. Immunosuppressive drugs such as cyclosporin A and FK506 inhibit NF-AT activation by binding to calcineurin subunits through immunophilin complexes, blocking substrate access to the catalytic domain.
