Target Information
TNF alpha is a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers of two, three, or five noncovalently linked units, but shows a single 17 kDa band under non-reducing SDS-PAGE conditions. Knockout studies in mice suggest a neuroprotective function, and TNF alpha induces tumor necrosis when injected into tumor-bearing mice.
TNF alpha plays roles in immune responses to bacterial, viral, parasitic, and fungal infections, and in the necrosis of specific tumors. It acts synergistically with interferon-gamma in cytotoxicity and is implicated in autoimmune diseases, insulin resistance, and cancer.
