Product Specific Information
This SRBCZX monoclonal antibody recognizes signal transducer and activator of transcription 5 (STAT5) when phosphorylated on tyrosine 694. STAT proteins are activated by ligand binding to receptors associated with Janus kinase (JAK) family members. Upon phosphorylation by JAKs, STAT proteins translocate to the nucleus to regulate gene transcription in a cell type- and cytokine-specific manner.
Cytokines such as IL-2, IL-7, and IL-15 (via the common gamma chain) induce STAT5 phosphorylation on Tyr694 by JAK1 and JAK3, while IL-3, IL-5, and GM-CSF (via the common beta chain) induce phosphorylation by JAK2. Phosphorylation at Tyr694 is essential for STAT5 dimer formation, nuclear translocation, and DNA binding.
Specificity of this SRBCZX clone was determined by ELISA and flow cytometry.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining of stimulated human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per 100 µL test volume (10⁵–10⁸ cells/test).
Staining Protocol: Use Protocol C (Two-step: Fixation/Methanol). Protocols A and B are not suitable.

Target Information
STAT5 alpha is a member of the STAT family of transcription factors. Upon cytokine or growth factor stimulation, STAT5 is phosphorylated by receptor-associated kinases and forms dimers that translocate to the nucleus to act as transcription activators. It mediates responses to IL-2, IL-3, IL-7, GM-CSF, erythropoietin, thrombopoietin, and various growth hormones.
Activation of STAT5 alpha has been linked to tumorigenesis in myeloma and lymphoma associated with TEL/JAK2 fusion. In mouse, STAT5 alpha induces expression of anti-apoptotic BCL2L1/BCL-X(L). STAT5 alpha and beta share 96% sequence similarity and are co-expressed in most tissues, though expression ratios vary by tissue type. STAT5 alpha is involved in reproduction, lactation, immune function, and somatic growth.
