FOXO1, FOXO3, and FOXO4 belong to the forkhead family of transcription factors.
- FOXO1 (FKHR): 70 kDa 단백질로, 핵 수용체 활성의 공활성자(coactivator) 또는 공동억제자(corepressor)로 작용할 수 있습니다. 이는 FOXO1A 단백질의 C-말단에 있는 LXXLL 모티프를 통해 매개됩니다.
- FOXO3: 세포사멸(apoptosis) 유도에 관여하며, MLL 유전자와의 전위는 2차 급성 백혈병과 관련이 있습니다.
- FOXO4: 인슐린 신호전달 경로 조절에 관여하며, IRE(Insulin-Response Elements)에 결합하여 IGFBP1 전사를 활성화합니다. 또한 HIF1A 발현을 억제하고 저산소 유도 전사 활성화를 감소시킵니다.
