Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. AR acts as a ligand-activated transcription factor binding to testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, AR translocates into the nucleus, dimerizes, and activates transcription of androgen-responsive genes.

AR signaling is essential for the development and function of male reproductive organs such as the prostate and epididymis, and also influences non-reproductive tissues including muscle, hair follicles, and brain. As a phosphoprotein, AR regulates MAP kinase pathways, and its phosphorylation state is linked to MEK1/2 activity. Dysregulation of AR signaling is associated with diseases such as prostate cancer, Kennedy’s disease, and male infertility. Mutations in the AR gene can cause complete androgen insensitivity syndrome (CAIS).
