Target Information
CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven transmembrane domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and also mediates HIV-2 binding through its envelope glycoprotein gp120, promoting viral fusion.
CXCR4 serves as the receptor for the chemokine CXCL12/SDF-1, which triggers intracellular calcium increase and MAPK1/MAPK3 activation. It also acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP signaling.
This receptor is involved in hematopoiesis, cardiac septum formation, and vascular development in the gastrointestinal tract. In the CNS, it supports hippocampal neuron survival and cerebellar development. CXCR4 also mediates LPS-induced inflammatory responses, including TNF secretion by monocytes.
Antibodies against CXCR4 can block HIV-1 and HIV-2 fusion and infection. The amino-terminal domain and the second extracellular loop of CXCR4 are essential for HIV binding.
