Target Information
CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven membrane-spanning domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and also mediates HIV-2 binding and fusion. CXCR4 serves as the receptor for the chemokine CXCL12/SDF-1, triggering intracellular calcium increase and MAPK1/MAPK3 activation. It also acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP signaling. CXCR4 plays roles in hematopoiesis, cardiac and vascular development, and neuronal survival. Antibodies to CXCR4 can block HIV-1 and HIV-2 infection of human target cells. The N-terminal domain and second extracellular loop are key HIV binding sites.
