Target Information

CXCR4 (also known as Fusin, LESTR, HUMSTR) is a member of the G-protein-coupled chemokine receptor family with seven membrane-spanning domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and has been shown to serve as a co-receptor for HIV-2 binding to CD4 through the envelope glycoprotein gp120, promoting Env-mediated fusion.

CXCR4 is also the receptor for the C-X-C chemokine CXCL12/SDF-1, mediating intracellular calcium increase and MAPK1/MAPK3 activation. It acts as a receptor for extracellular ubiquitin, leading to enhanced intracellular calcium and reduced cAMP levels. CXCR4 is involved in hematopoiesis, cardiac ventricular septum formation, and vascularization of the gastrointestinal tract, likely through regulation of endothelial cell branching and remodeling. In the CNS, CXCR4 mediates hippocampal neuron survival and cerebellar development, and also binds bacterial lipopolysaccharide (LPS), mediating LPS-induced inflammatory responses including TNF secretion by monocytes.

Antibodies to CXCR4 can block HIV-1 and HIV-2 fusion and infection of human target cells. The amino-terminal domain and the second extracellular loop of CXCR4 serve as HIV binding sites.
