이 항체는 ZO-1 단백질과 반응하며, 서열 상동성에 근거할 때 ZO-2 또는 ZO-3과의 반응은 예상되지 않습니다.
Immunofluorescence 및 Immunohistochemistry에서 성공적으로 사용되었습니다. Western blotting의 경우 monoclonal anti-ZO-1 antibody (Cat. No. 339100) 또는 다른 polyclonal ZO-1 antibody (Cat. Nos. 617300, 402200)를 권장합니다.
- MDCK-II, Caco-2, Rat-1, NRK52E 세포에서 Immunofluorescence로 반응 확인
- Frozen mouse liver, heart blood vessel, brain mossy fiber terminal 조직에서 IHC/IF 반응 확인
- Formalin-fixed, paraffin-embedded human pancreas, prostate, small intestine 조직에서 IHC 반응 확인
- Formalin-fixed, paraffin-embedded 조직의 경우, 염색 전 pepsin 전처리 필요
- 일부 lot은 human, dog, rat cell lysate를 이용한 Western blotting에서도 사용됨
- Immunofluorescence에서는 methanol fixation 권장 (paraformaldehyde fixation도 가능)
Target Information
ZO-1은 ZO-2 또는 ZO-3과 복합체를 형성하며, claudin, occludin, F-actin과도 결합하여 tight junction에서 actin cytos켈레톤과 연결 역할을 합니다.
ZO-1 발현은 여러 유방암 세포주에서 현저히 감소되어 있으며, ZO-2와 ZO-3은 상피세포의 tight junction에 널리 발현됩니다.
ZO-1은 세포 간 tight junction의 세포질막 표면에 위치하며, 세포-세포 결합에서 신호전달에 관여할 수 있습니다.
ZO-1 유전자는 두 가지 isoform을 암호화하는 전사 변이체를 가지고 있습니다.
ZO-1 기능 이상은 Celiac Disease 및 Congenital Nephrotic Syndrome과 관련이 있습니다.
