Product Specific Information

The 104 monoclonal antibody reacts with the mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.2-expressing mouse strains such as C57BL/6, BALB/c, DBA, C3H, CBA, 129, A, and AKR. CD45.2 is expressed by all leukocytes in these strains.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocyte suspensions. Recommended ≤0.5 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best performance.

Fluorochrome Information:
Alexa Fluor® 700 emits at 723 nm and is excited with a red laser (633 nm). Recommended detection setup: 685 LP mirror and 710/20 filter.

Target Information

CD45.2 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It regulates Src-family kinases, influences lymphocyte development and antigen signaling, and exists in multiple isoforms due to alternative splicing. These isoforms differ in extracellular domains and membrane localization, depending on cell type and physiological state. CD45.2 also modulates integrin-mediated signaling and participates in apoptosis-related DNA fragmentation. CD45RA is an isoform with restricted expression among lymphoid subtypes.
