Reconstitution:
Lyophilized human IL-23 should be reconstituted in water to a protein concentration of 0.1–1.0 mg/mL. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein, such as 0.1% BSA, and store in working aliquots at -80°C to -20°C.
Storage:
Lyophilized human IL-23 should be stored at 2°C to 8°C, preferably desiccated. Store reconstituted human IL-23 at ≤-20°C (not in a frost-free freezer). Minimize freeze-thaw cycles.
Target Information
IL-23 is a heterodimeric cytokine composed of the p40 subunit of IL-12 disulfide-linked with a protein p19. The p19 subunit, like p35 of IL-12, is biologically inactive by itself. IL-23 interacts with IL-12Rβ1 and an additional, novel β2-like receptor subunit with a STAT4 binding domain, termed IL-23R.
IL-23 is secreted by activated mouse and human dendritic cells.
Biological activities of mouse IL-23 differ from those of IL-12:
- Mouse IL-23 does not induce significant IFN-γ production.
- It induces strong proliferation of memory T cells (but not naïve T cells).
- IL-23 activates memory T cells to produce proinflammatory cytokine IL-17.
Human IL-23 shows similar but distinct properties from IL-12:
- Induces proliferation of memory T cells.
- Induces moderate IFN-γ production by naïve and memory T cells compared to IL-12.
