Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ or CD8αα dimers. CD8 binds to MHC class I and, through association with protein tyrosine kinase p56lck, plays a role in T cell development and activation.

Applications Reported: Flow cytometric analysis of mouse splenocytes.
Applications Tested: ≤ 0.25 µg per test (100 µL final volume, 10⁵–10⁸ cells). Antibody titration recommended for optimal performance.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein involved in signal transduction and T cell differentiation. It forms a heterodimer (CD8αβ) acting as a co-receptor for MHC class I molecules, while CD4 serves as a co-receptor for MHC class II. CD8 is expressed on T cells, NK cells, monocytes, and dendritic cells. Engagement with MHC-I/peptide complexes recruits Lck kinase, leading to CTL activation and pathogen/tumor clearance. Differentiation of naive CD8+ T cells into CTLs is promoted by IL-2, IL-12, and TGF-β1.
