Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or starvation for pyrimidine nucleotides.
p53 up-regulates stress response genes such as p21waf1 and other proteins involved in G1 or G2/M arrest. Structurally, p53 contains an N-terminal transactivation domain, a central DNA-binding domain, an oligomerisation domain, and a C-terminal regulatory domain.
Phosphorylation sites, especially Ser15 and Ser392, are critical for activation, stabilization, and tumor association. p53 functions as a key anticancer agent by blocking proliferation of damaged cells.
It is expressed at low levels in normal cells but highly expressed in transformed cell lines, contributing to malignancy. Mutations in TP53 lead to loss of tumor suppressor activity and are associated with various human cancers, including Li-Fraumeni syndrome.
